Epitalon vs Pinealon: One Lab Built Both. Epitalon Has an FDA Review and Three Small Human Studies. Pinealon's Case Rests Mostly on Cell Dishes and Rats.
Epitalon vs pinealon is a comparison between two short synthetic peptides from the same research program in St. Petersburg, Russia. Epitalon is a four amino acid peptide, Ala-Glu-Asp-Gly (AEDG), designed from a pineal gland extract and studied mostly for melatonin, aging markers and lifespan in animals. Pinealon is a three amino acid peptide, Glu-Asp-Arg (EDR), studied mostly for nerve cell protection in cell cultures and rats. Despite the name, pinealon's published work is about the brain, not the pineal gland. The practical gap between them is the depth of the human evidence. Epitalon has three small human studies and a full FDA evaluation published for a July 2026 advisory committee meeting. Pinealon has a handful of small Russian-language clinical reports and no FDA review at all. Neither has a large, independent, placebo-controlled trial, and no published study has tested epitalon and pinealon together.
Key Takeaways: Epitalon vs Pinealon
Epitalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly (AEDG), built from the amino acid composition of epithalamin, a bovine pineal gland extract, according to a 2025 review by Araj and colleagues in the International Journal of Molecular Sciences. Pinealon is the synthetic tripeptide Glu-Asp-Arg (EDR), described by Khavinson and colleagues in Rejuvenation Research in 2011 as limiting the build-up of reactive oxygen species in nerve and immune cells grown in the lab. Epitalon's sleep-related evidence is about melatonin: in a 20-day study of 40 night-shift women with low melatonin output, a sublingual AEDG spray raised the urinary melatonin metabolite 1.7-fold, as summarized in FDA's 2026 briefing. Pinealon has no comparable sleep or melatonin study in the PubMed record we searched. Pinealon's strongest signal is in brain cell models: in mouse hippocampal neurons exposed to amyloid, EDR at 200 ng/ml raised mature dendritic spines by 71% (Kraskovskaya and colleagues, 2017). That is a cell culture result, not a human outcome. The FDA concluded in 2026 that there are no clinical data to support the safety of epitalon in humans. Pinealon does not appear in that briefing or among the peptides the advisory committee considered in July 2026.
What is the difference between epitalon and pinealon?
The core difference between epitalon and pinealon is structure and target. Epitalon has four amino acids (alanine, glutamic acid, aspartic acid, glycine) and was designed to imitate a pineal extract. Pinealon has three (glutamic acid, aspartic acid, arginine) and has been studied as a protector of nerve cells. Epitalon has a defined chemistry record at the FDA. The agency's 2026 evaluation gives the free base the formula C14H22N4O9 and a molecular weight of 390.35 g/mol, and states that the peptide was first synthesized in the late 1980s by researchers working under Professor Vladimir Khavinson. Pinealon comes from the same circle: nearly every pinealon paper in PubMed lists Khavinson or the St. Petersburg Institute of Bioregulation and Gerontology among its authors or affiliations. The shared origin matters for how much weight the evidence can carry. When almost all the studies on a compound come from the group that developed it, independent replication is the missing step, and for pinealon that step has barely started.
Does pinealon act on the pineal gland like epitalon?
Pinealon does not appear to act on the pineal gland the way epitalon does, at least in the one study that compared them directly in pineal tissue. The name suggests a pineal peptide, but the published work on pinealon centres on the brain cortex, the cerebellum and the hippocampus. Khavinson and colleagues, writing in the Bulletin of Experimental Biology and Medicine in 2011, grew pineal gland tissue from 3-month-old rats and exposed it to several short peptides. AEDG, the epitalon sequence, raised the proliferation marker Ki-67, and AEDG was the only peptide that stimulated pinealocytes to produce the signalling protein CGRP. Glu-Asp-Arg, the pinealon sequence, did not change the cell death marker AIF, and the authors concluded that AEDG acts on pineal cells in a tissue-specific way. That is a single rat tissue study, so it cannot settle what either peptide does in a person. It does undercut the assumption, common on vendor pages, that pinealon and epitalon are interchangeable pineal peptides.
Epitalon vs pinealon for sleep: which has better evidence?
For sleep, epitalon has the better evidence, though it is thin. The data concern melatonin, the hormone that signals night to the body, rather than measured sleep quality. The most cited epitalon study is Ivko and colleagues, 2021, summarized in FDA's 2026 briefing. Researchers screened 75 healthy women aged 40 to 50 working night shifts and found 40 with low output of 6-sulfatoxymelatonin, the main urinary breakdown product of melatonin. Those 40 received either a placebo spray or an AEDG spray under the tongue for 20 days, delivering 0.5 mg of peptide a day. On AEDG, the melatonin metabolite rose 1.7-fold; on placebo it did not change. The authors reported improved psycho-emotional and physical condition in 83% of the AEDG group against 25% on placebo, but the FDA noted that the publication did not specify blinding or describe how that condition was measured. Animal data point the same way. Goncharova and colleagues, Experimental Gerontology, 2005, reported that epitalon raised night melatonin in old rhesus monkeys aged 20 to 27 years but had no effect in young animals aged 6 to 8. A 2007 Russian-language report by Korkushko and colleagues described a rise in night melatonin in elderly people with low pineal function on epitalon or the pineal extract epithalamin. Pinealon has no equivalent. We found no published pinealon study that measured melatonin, sleep duration or sleep quality in people. Searches for epitalon vs pinealon for sleep are, in practice, a search for epitalon.
Epitalon's Melatonin Study in Night-Shift Women
Percent of women reported to show improved psycho-emotional and physical condition after 20 days of sublingual AEDG (0.5 mg a day) or placebo. Urinary melatonin metabolite rose 1.7-fold on AEDG. Blinding was not specified and the measure was not described (Ivko and colleagues, 2021, as summarized in FDA's 2026 briefing). No comparable pinealon study exists.
What does the research show pinealon does in the brain?
Pinealon's research shows nerve cell protection in lab and animal models, mostly from the Khavinson group. The results are consistent in direction but have not been tested in a controlled human trial. In cell cultures, Khavinson and colleagues reported in Rejuvenation Research in 2011 that pinealon reduced reactive oxygen species in a dose-dependent way in rat cerebellar granule cells, immune cells called neutrophils, and PC12 nerve-like cells, and that it reduced necrotic cell death. A 2014 paper in the Bulletin of Experimental Biology and Medicine reported that Glu-Asp-Arg stimulated serotonin expression in aging brain cortex cultures. The most striking number comes from an Alzheimer's disease model. Kraskovskaya and colleagues, Bulletin of Experimental Biology and Medicine, 2017, exposed mouse hippocampal neurons to toxic amyloid and then added EDR at 200 ng/ml. The count of mushroom spines, the mature connection points between neurons, rose by 71% and returned to normal. A related peptide, KED, at the same concentration raised spines by 20%. A 2021 follow-up in Pharmaceuticals reported that EDR and KED prevented spine loss in 5xFAD mice, a strain bred to develop Alzheimer's-like plaques. In rats, Arutjunyan and colleagues, International Journal of Clinical and Experimental Medicine, 2012, gave pinealon to pregnant rats fed excess methionine and reported better learning and spatial orientation in the offspring. A 2015 study in 18-month-old rats under low oxygen and mild cold found that a comparison product, Cortexin, had a stronger effect than pinealon on free radical processes and the cell death enzyme caspase 3.
Pinealon's Strongest Signal Is in a Dish
Increase in mushroom spines in mouse hippocampal neurons exposed to amyloid, with each tripeptide at 200 ng/ml. EDR returned spine counts to normal. Cell culture only (Kraskovskaya and colleagues, Bulletin of Experimental Biology and Medicine, 2017).
Has pinealon been studied in humans?
Pinealon has been studied in humans only in a few small reports, published in Russian, and usually in combination with other peptides. None is a large randomized placebo-controlled trial. Meshchaninov and colleagues, Advances in Gerontology, 2015, gave pinealon and a second tripeptide, vesugen, to 32 people aged 41 to 83 with several chronic conditions and organic brain syndrome in remission. The authors reported slower aging on biological age markers, and judged vesugen the more effective of the two. They also reported pro-oxidant activity and a drop in CD34-positive blood-forming stem cells, which they described as inhibition of blood cell production. Those are findings a reader weighing pinealon should know about, and the paper itself says the question needs more study. Bashkireva and Artamonova, Advances in Gerontology, 2012, examined 150 male truck drivers with a mean age of 43.3 and 150 metal workers. They reported that bioregulating peptides improved psycho-emotional measures in drivers, with the best result from pinealon combined with vezugen. The English abstract does not give the number of men who received peptides or the size of the effect. A 2020 review by Khavinson and colleagues in Molecules states that EDR improves memory issues in elderly patients, citing the group's own earlier work. Independent confirmation of that claim has not been published in the PubMed record we searched.
How Many People the Human Studies Enrolled
Participants in the human studies discussed in this article. Ivko 2021: 40 night-shift women with low melatonin output split between AEDG and placebo (FDA briefing, 2026). Meshchaninov 2015: 32 people aged 41 to 83 given pinealon and vesugen. Bashkireva and Artamonova 2012: 150 male truck drivers examined; the abstract does not say how many received peptides.
What has epitalon been shown to do in animals and people?
Epitalon has a longer animal record than pinealon, with mixed lifespan results and a small human evidence base. The key mouse study is Anisimov and colleagues, Biogerontology, 2003. Female SHR mice, 54 per group, received 1.0 microgram of Epitalon on 5 consecutive days each month from 3 months of age. Mean lifespan did not change. The last 10% of survivors lived 13.3% longer, maximum lifespan rose 12.3%, chromosome aberrations in bone marrow fell 17.1%, and leukemia was inhibited 6.0-fold. The most quoted human figures do not belong to synthetic epitalon. Khavinson and Morozov, Neuro Endocrinology Letters, 2003, followed 266 older people given thymalin, epithalamin or both over 6 to 8 years, and reported 1.6 to 1.8-fold lower mortality in the epithalamin-treated group. Epithalamin is the pineal extract. That was a different substance, and the FDA treats epithalamin and epitalon as distinct. For synthetic epitalon itself, the FDA found three human studies: two sublingual studies in night-shift workers and one eye-injection study in retinitis pigmentosa. Reviewers found no pharmacokinetic data, meaning nobody has published how the peptide is absorbed, distributed or cleared in people.

Can you take epitalon and pinealon together?
No published study has tested epitalon and pinealon together, so the safety and effect of an epitalon and pinealon stack are unknown. Online protocols that pair them are extrapolating from separate, small studies. The Khavinson group has tested combinations of its own peptides, but not this pair. Khavinson and Morozov's 2003 report combined the pineal extract epithalamin with the thymus extract thymalin and reported a 2.5-fold lower mortality than controls. The truck driver report paired pinealon with vezugen. Neither result says anything about combining synthetic epitalon with pinealon. There is also no human dosing data for pinealon in the abstracts we reviewed, and no pharmacokinetic data for epitalon according to the FDA. Without either, any combined dose is a guess. Anyone considering these peptides should talk to a physician who can review the full medication list and order baseline labs.
What is the regulatory status of epitalon and pinealon?
Neither epitalon nor pinealon is an FDA-approved drug. Epitalon went through a formal FDA review in 2026; pinealon has not been reviewed. For the July 23 to 24, 2026 meeting of the Pharmacy Compounding Advisory Committee, FDA staff evaluated epitalon and proposed not adding it to the 503A Bulks List, the list of substances pharmacies may use for compounding. The agency concluded that there are no clinical data to support epitalon's safety in humans, and raised a theoretical concern that long-term telomerase activation could let cells evade senescence and become cancerous. The committee disagreed with staff and voted 7-5, with one abstention, to recommend epitalon for insomnia, according to RAPS. The final decision rests with the FDA. The same committee voted 6-7 against emideltide, another peptide proposed for insomnia. Pinealon is not mentioned in FDA's epitalon briefing and was not among the peptides the committee considered.

Which should a man researching epitalon vs pinealon look at first?
A man comparing epitalon vs pinealon should look first at what each peptide was actually studied for, because the two are not substitutes. Epitalon's data concern melatonin, aging markers and lifespan in animals, plus a 20-day melatonin study in 40 women. Pinealon's data concern nerve cell survival, oxidative stress and dendritic spines, almost entirely in cells and rats. Both records share the same weakness: nearly every study comes from the group that created the peptides, the human studies are small, and the most important safety questions, such as long-term effects and how the body handles the peptides, are unanswered. For epitalon, the FDA said so directly. For pinealon, no regulator has looked. That does not make either peptide useless as a research subject. It means the claims attached to them online, from deeper sleep to sharper memory, are running well ahead of the trials.
