Peptides & Longevity

The Entire Case for Magnesium and Testosterone Rests on Thirty Men in Turkey, Four Weeks, and a Dose Twice the Supplemental Upper Limit. The Ten Who Took No Magnesium Went Up Too.

Magnesium appears in almost every testosterone stack on the market, and the number behind that placement comes from one 2011 study by Cinar and colleagues: thirty men aged 18 to 22, split into three groups of ten, four weeks, 10 mg per kilogram of body weight a day, which is about 800 mg for an 80 kg man against a 350 mg upper limit for supplements. Total and free testosterone rose in all three groups, including the ten athletes who took no magnesium at all. Peptides & Longevity

One Week of Five-Hour Nights Cut Daytime Testosterone by 10 to 15 Percent in Ten Healthy Young Men, a Drop That Normal Aging Takes 10 to 15 Years to Produce. Here Is What Every Human Sleep and Testosterone Study Actually Found.

Sleep is the only testosterone intervention with a tightly controlled human experiment behind it. In the 2011 JAMA study by Leproult and Van Cauter, ten healthy men aged about 24 slept five hours a night for eight nights in a laboratory and their daytime testosterone fell 10 to 15 percent, with the deepest trough between 2 pm and 10 pm. Older men who slept around four hours had morning testosterone roughly half that of men sleeping eight, and in 2,295 US men each hour of sleep lost tracked with about 5.85 ng/dL less testosterone. This article walks through every controlled restriction study, explains why a single 8 am blood draw can miss the effect, and separates the sleep signal from the obesity and sleep apnea it travels with.

Peptides & Longevity

Zinc Restriction Cut Testosterone by 73 Percent in Four Healthy Men, and 30 mg a Day Nearly Doubled It in Nine Deficient Older Men. In Men Who Already Get Enough, Three Trials Found Nothing, and Supplement Doses Over 100 mg Doubled the Risk of Advanced Prostate Cancer in 46,974 Men.

Zinc has the strongest deprivation evidence of any testosterone supplement: a 1996 study that fed four healthy men a zinc-restricted diet for 20 weeks saw serum testosterone fall from about 1,150 to 306 ng/dL, and 30 mg a day for six months raised testosterone from 239 to 461 ng/dL in nine marginally deficient older men. The only double-blind placebo-controlled trial, 20 men on hemodialysis given 50 mg for six months, found testosterone rose from 280 to 520 ng/dL. But in men who already eat 12 to 23 mg a day, ZMA at 30 mg did nothing to total or free testosterone in three studies totalling 83 men. The tolerable upper limit is 40 mg, doses of 50 mg or more deplete copper over months, and men taking more than 100 mg a day had a 2.29 times higher rate of advanced prostate cancer in a 46,974-man cohort.

Tongkat Ali Raised Total Testosterone by 24 ng/dL in 105 Men Over 50 and by Nothing in 149 Healthy Men. The Meta-Analysis That Rates It Best Found 87 Percent Heterogeneity, and Europe's Food Safety Regulator Concluded Its Safety Has Not Been Established.

Tongkat ali (Eurycoma longifolia) has nine human studies that measured testosterone in men, seven of them funded or co-authored by the maker of the Physta extract. The largest randomized trial, 105 men aged 50 to 70 with testosterone under 300 ng/dL, found total testosterone rose from 200.5 to 225.0 ng/dL on 200 mg a day over 12 weeks while placebo fell from 183.0 to 177.9. Two trials in 149 men with normal testosterone found no difference from placebo. A 2022 meta-analysis of five randomized trials and 232 men reported a standardized mean difference of 1.35 with 87 percent heterogeneity and a positive publication-bias test. In December 2021 the European Food Safety Authority concluded the extract's safety had not been established under any condition of use after a positive chromosome aberration test and a positive in vivo comet assay. Peptides & Longevity

Spermidine Has the Cleanest Mechanism in the Longevity Aisle. The Only Year-Long Human Trial Raised the Dose by Ten Percent and Found Nothing.

Spermidine extends lifespan in yeast, flies, worms and mice, and its cardiac benefit vanishes in mice lacking the autophagy gene Atg5. The Bruneck cohort of 829 adults linked higher dietary spermidine to a hazard ratio of 0.76 for all-cause death per standard deviation of intake. SmartAge, the only 12-month randomized placebo-controlled trial, used 0.9 mg per day and missed its primary endpoint with a between-group difference of minus 0.03. An educational review of what the spermidine evidence actually shows for men.

One Gram of Fish Oil and the Clock That Moved
Three Years, 2,157 People, Six Primary Endpoints That All Came Back Null, and a Post Hoc Methylation Analysis in 777 of Them That Became the Headline
Peptides & Longevity

One Gram of Fish Oil a Day Slowed Biological Aging by Three to Four Months in a Randomized Trial. The Same Trial Missed All Six of the Endpoints It Was Built to Answer.

The DO-HEALTH trial randomized 2,157 adults aged 70 and older to vitamin D, marine omega-3 and home strength exercise for three years and reported no benefit on any of its six primary endpoints. A post hoc analysis of 777 participants published in Nature Aging in February 2025 found that 1 gram of omega-3 per day slowed three of six DNA methylation clocks, with effects of 0.16 to 0.32 standardized units, or roughly 2.9 to 3.8 months. An educational review of what the omega-3 biological aging evidence actually shows for men.

The Only Randomized Caloric Restriction Trial in Humans Slowed the Pace of Aging by Three Percent. It Also Took Two Percent Off the Hip.

CALERIE Phase 2 randomized 220 healthy non-obese adults to two years of 25 percent caloric restriction or normal eating. Participants delivered 11.9 percent. Cardiometabolic risk fell sharply, one epigenetic clock out of six slowed by about 3 percent, bone density dropped roughly 2 percent at the spine and femoral neck, and free testosterone dipped at month 12 before recovering. An educational review of what the human caloric restriction evidence actually shows for men.

A Twelve Percent Male Lifespan Gain, a Second Cohort That Found Nothing, and a Dose Gap of Roughly a Hundred Fold Between the Mouse Diet and the Capsule
Peptides & Longevity

Astaxanthin Added 12 Percent to Male Mouse Lifespan. The Same Program Ran It Again and Got Nothing.

The NIA Interventions Testing Program reported that astaxanthin extended male mouse median lifespan by 12 percent at p equals 0.003, with no effect in females. In 2026 the same program tested astaxanthin a second time and found no lifespan effect in either sex. The mice were eating a human-equivalent dose near 1,600 milligrams a day. Sixty Sessions of Hyperbaric Oxygen Lengthened Telomeres by a Third. There Were Twenty-Five People and No Control Group.

Hachmo and colleagues reported in Aging in 2020 that 60 daily hyperbaric oxygen sessions raised average telomere length in blood cells by 20 to 38 percent in adults over 64, alongside a 37 percent drop in senescent T helper cells. The telomere analysis included 25 people, there was no control arm, and clearing short-telomere cells raises the average without lengthening anything. Acarbose Added 22 Percent to Male Mouse Lifespan. The Five-Year Human Trial Was Measuring the Heart.

Acarbose raised male mouse median lifespan by about 22 percent in the National Institute on Aging Interventions Testing Program, replicated across three laboratories, against roughly 5 percent in females. Start the same dose at 16 months instead of 4 and the male gain falls to about 6 percent. In humans it is an approved diabetes drug with two large randomized trials, neither of which measured aging. 17-alpha-estradiol raised male mouse median lifespan by roughly 19 percent in the National Institute on Aging Interventions Testing Program, replicated across three laboratories, and it still worked when treatment started at 16 months of age. Female mice gained nothing at any dose. Males castrated before treatment lost nearly the entire response. Alpha-Ketoglutarate and the Aging Question
A Striking Mouse Result, an Eight-Year Age Reversal With Nothing to Compare It To, and Two Failures at the Program Built to Check
Peptides & Longevity

Alpha-Ketoglutarate's Eight-Year Age Reversal Came From 42 Customers and No Control Group. The Program That Runs Controls Just Reported Nothing.

Alpha-ketoglutarate became a longevity staple on the strength of two results: a 2020 mouse study in which frailty scores fell 46 percent, and a 2021 paper reporting an average eight-year drop in DNA methylation age among 42 people taking an AKG supplement. The second study had no placebo group, no randomization and no comparison group at all, and the participants were customers who had already bought the product. In 2026 the National Institute on Aging Interventions Testing Program reported that alpha-ketoglutarate did not extend lifespan in genetically heterogeneous mice, started early or late. Canagliflozin is the rare geroprotector with a clean mouse lifespan result: 14 percent added to male median survival across three independent test sites, repeated when the drug was started at 16 months of age, and absent in females both times. The human record is enormous and points somewhere else entirely, because all of it was collected in people who already had heart disease, heart failure or kidney disease. Metformin's Longevity Trial Has Not Enrolled a Single Person. The Exercise Trials Ran Anyway, and They Keep Finding the Same Thing.

Metformin became the default longevity drug on the strength of a database study in which 78,241 diabetics outlived 90,463 matched people without diabetes. The trial designed to test that properly, TAME, has been ready since 2015 and still has zero participants. Meanwhile three randomized trials added metformin to supervised exercise and watched the training adaptation shrink: lean mass gains fell from 1.95 percent to 0.41 percent, VO2 max gains disappeared, and mitochondrial respiration stopped responding.
